Building basement room over gravel/vapor barrier?

Yeah, what you describe so far sounds good. Rot and mold grows with moisture so eliminate it. Make sure the outside of the stucture is well drained. Slope all outside earth away from the foundation so water does not collect along the walls. Depending on your area and aount of moisture you can also install water proofing on the outside if required.
